Scientists Report Key Finding in Breast Cancer Research

Researchers at Drexel University College of Medicine have discovered that a certain type of sugar found naturally in the body is elevated in breast cancer cells and is critical for cancer growth and movement. When researchers reduced and normalized the levels of this sugar, they were able to slow the growth of the cancer cells and block invasion. The findings, published in the March 1 issue of Oncogene, represent a potential new therapeutic target for treating aggressive forms of breast cancer.

Scientists have known for years that cancer cells are addicted to sugar. Compared to neighboring normal cells, cancer cells take up nearly ten times more sugar. The cancer cells use the increased sugar levels to fuel rapid cell growth and spread. The Drexel researchers studied a particular sugar-based protein modification known as O-GlcNAc. Alterations in this modification have been linked previously to diabetes and Alzheimer?s disease, but not to cancer.

?This sugar, O-GlcNAc, is used inside cells to tag proteins and alter their function,? said lead author Mauricio J. Reginato, Ph.D., assistant professor in the Department of Biochemistry and Molecular Biology at Drexel University College of Medicine. ?We think this is a very exciting discovery because this may be one way that cancer cells use the increased sugar to regulate key pathways that control growth and invasion.?

Reginato and his colleagues studied established cells from patients with estrogen-independent breast cancer, the most aggressive form of the disease, which does not respond well to current treatments. The researchers found that compared to normal mammary cells, all the breast cancer cells contained elevated levels of O-GlcNAc, as well as increased levels of the enzyme responsible for adding this sugar to protein.

When they reduced the levels of the O-GlcNAc enzyme, or blocked its activity with a drug, they reduced the growth of breast cancer and also blocked the invasion. In addition, by reducing the enzyme levels, they also reduced levels of a protein known as FoxM1, which is known to play a critical role in controlling cancer growth and spread.

The Drexel researchers and collaborators are now working to develop more potent chemicals to target the O-GlcNAc enzyme, a potential new therapeutic target for treating breast cancer and possibly other cancers.

?We looked at cancer databases and found that this enzyme is also elevated in other types of cancers.? said Reginato. ?Our next step will be to study those cancers more closely to determine if we can achieve the same effect.?

American Association of Poison Control Centers Releases Annual Poison Exposure Report

America?s 57 poison centers fielded 3.9 million calls in 2010, an average of nearly 11,000 per day, according to the recently published 2010 Annual Report of the American Association of Poison Control Centers? National Poison Data System. The 28th annual report was published in the December 2011 issue of Clinical Toxicology.

The American Association of Poison Control Centers (AAPCC) maintains the National Poison Data System (NPDS), a large database containing information about all poison exposure phone calls to every poison center across the country. NPDS is the only near real-time, comprehensive poisoning surveillance database in the United States.

According to the report, poison experts at the nation?s poison centers treated 2.4 million human poison exposures and handled 1.5 million information calls in 2010. Children younger than 6 accounted for about half of all the poison exposure calls; however, adults 20 and older accounted for 92 percent of all deaths reported.

A total of 1,730 deaths were reported in 2010; of those, 139 were younger than 20, including 55 younger than 6. Sedatives/hypnotics/antipsychotics, cardiovascular drugs, opioids, and acetaminophen combinations were most frequently associated with poison-related deaths.

Poison exposures with serious outcomes increased 4.5 percent over the previous year. Eighty-one percent of poisonings were unintentional, and 15 percent were intentional. Of the 15 percent, 9 percent were suspected suicides.

?Poisonings continue to be a significant cause of illness and death in the U.S.,? said Alvin C. Bronstein, M.D., lead author of the report. ?The NPDS provides a nationwide infrastructure for public health surveillance of poison exposures, including drug, consumer product, environmental and food poisoning. This report supports the value of poison centers and the need for specialized medical toxicology expertise across the country.?

The 2010 report also highlights the vital role of poison centers in the nation?s public health system. For example, poison centers tracked the health impact of the 2010 Gulf of Mexico oil spill and raised the alarm about the emerging public health threats of designer amphetamines (Bath Salts) and designer marijuana (Spice and variants).

?The ability of the NPDS to track emerging public health threats makes it a valuable, vital, model system for national and global public health,? said Richard C. Dart, M.D., Ph.D., AAPCC president. ?When someone calls a poison center, information about that poison exposure is uploaded to the NPDS within 20 minutes. Because the data is so current, poison centers are often the first to identify contaminated food outbreaks, tainted medication, drug abuse threats or other emerging public health crises.?

Other findings in the report include:

About 75 percent of all calls to poison centers originated in people?s homes.
About 71 percent of the 2.4 million people who called with poison emergencies were treated at home, saving millions of dollars in medical expenses.
The top five substances most frequently involved in human poisonings were analgesics (11.5%), cosmetics/personal care products (7.7%), household cleaning substances (7.3%), sedatives/hypnotics/antipsychotics (6%) and foreign bodies/toys/miscellaneous (4.2%).

Alzheimer’s Disease Cutting Edge Research Announced on Natural Health News Report Website

Dr. Marambaud and his colleagues also tested a handful of other antioxidants against beta-amyloid. Only Resveratrol stood out. In addition, he adds that Resveratrol has been shown to have some “very interesting pharmacological effects” against herpes, some cancers, and possibly neurodegenerative diseases such as Huntington’s.

Marambaud’s interest was sparked by observational studies (done by other experts) showing that Alzheimer’s is rarer in populations that consume moderate amounts of red wine.

In fact, in France for example, many people are living longer lives despite a diet high in saturated fat. This phenomenon has been labeled the “French Paradox.” Researchers believe that red wine intake can help prevent certain Cancers, heart disease and Alzheimer’s disease.
